The Role of Glutamatergic Neurotransmission in Pathophysiology of Schizophrenia
Glutamátergní neurotransmise a její rolev patofyziologii schizofrenie
Poruchy funkce neuronů obsahujících excitační aminokyseliny se uplatňujív patofyziologii širokého spektra neurodegenerativních chorob. Dysfunkce neurotransmisezprostředkované excitačními aminokyselinami se může podílet také na patogenezi schizofrenie.Existují dvě glutamátové teorie patofyziologie schizofrenie. Hypotéza glutamátergní hypofunkcepředpokládá hypofunkci mozkových glutamátergních neuronů. Nejvýznamnějším důsledkemtéto hypotézy je možné terapeutické ovlivnění schizofrenní symptomatiky farmaky zvyšujícímiglutamátergní neurotransmisi. Byla zformulována také hypotéza glutamátergní hyperfunkce,jejímž východiskem je možné ovlivnění schizofrenní symptomatiky látkami snižujícímivyplavování glutamátu.

The dysfunction of the neurons containing excitatory amino acids is present in a wide range ofneurodegenerative disorders. Schizophrenia may also result from a dysfunction of excitatoryamino acid neurotransmission. There are two glutamatergic hypotheses of schizophrenia.Glutamate hypofunction hypothesis supposes hypofunction of cerebral glutamatergic neuronsin schizophrenia. The most signifi cant consequence of the hypothesis is a possible use of drugsenhancing glutamatergic neurotransmission in the treatment of schizophrenic symptoms.Glutamate hyperfunction hypothesis was also proposed. It suggests a possible effect of glutamaterelease-inhibiting drugs in schizophrenia.
